Mudbox's 3D Layers behave much like 2D layer systems of mainstream image editors such as Photoshop®. When sculpting on a layer, Mudbox calculates the difference between the positions of vertices after a brush stroke and their original positions on the "base" mesh.

Lights illuminate the objects in your scene and help you accurately evaluate the form of your sculpted mesh. You can modify the brightness (intensity) and color properties (diffuse and specular) for a light. A default light always exists for the 3D View in Mudbox, but additional lights can be created.
